Abstract | Dunaliella parva grew in different nitrogen concentrations, which were associated with lipid accumulation. D. parva could accumulate lipids. However, the underlying mechanism of lipid degradation requires further research. Thus, it is important to analyze lipid degradation to elucidate lipid accumulation. In this report, a gene encoding lipase from the green alga D. parva was isolated and characterized. Experimental results showed that the full-length cDNA of lipase gene was 4,583 bp, with an open reading frame of 3,183 bp, a 73 bp 5'-untranslated region and a 1,327 bp 3'-untranslated region. Potential regulatory elements related to hormones, light and unfavorable environments were found in the 1,828 bp 5'-flanking region. Analysis of lipase gene expression showed a correlation between lipase gene expression and nitrogen concentrations, which indicated that lipase gene may be responsive to nitrogen limitation stress. The cloning and expression analysis of lipase gene provided a basis for studying the regulatory mechanism of lipid degradation and the massive accumulation of lipids or lipase in D. parva. |
